| In vitro study | BAM7 binds directly to the BH3 structural binding groove at the N-terminus of BAX. BAM7 interacts with BAX directly on the surface, triggering BAX activation through the BIM BH3 helix. BAM7 leads to functional BAX activation. BAM7 triggers the switch of BAX from monomer to oligomer, this effect is dose and time-dependent, and the kinetics of BAX:BAM7 approaches the saturation 1:8 dose ratio. In vitro, BAM7 triggers BAX oligomerization, BAX-mediated pore formation, and BAX-dependent cell death. BAM7 selectively induces BAX-mediated apoptosis by triggering intracellular BAX activation. BAM7 kills only BAX-containing cell lines, producing biochemical and morphological features of BAX-mediated apoptosis. |
